A multi-well based detection platform with integrated PDMS concentrators for rapid multiplexed enzymatic assays

Abstract

We report an Integrated System for Accelerating Assays with Concentrators in a standard 12-well plate (ISAAC-12) and demonstrate its versatility in the matrix metalloproteinase (MMP)-9 assay by accelerating its enzymatic reaction via electrokinetic concentration. Based on Michaelis-Menten Equation, the reaction rate of enzymatic assays is dependent on substrate-enzyme concentration. Instead of the de facto laboratory standard platform multi-well plates, ISAAC can be easily applied to enzymatic assays for rapid multiplexed detection by concentrating the substrate-enzyme and/or the reaction product inside each microwell simultaneously in a highly parallel fashion.
