Abstract

(Chemical Equation Presented) Changing its nature: Global incorporation of noncanonical amino acids can alter the behavior of proteins in useful ways. In some cases, however, replacement of natural amino acids by noncanonical analogues (blue bars in picture) can cause loss of protein stability. After several generations, functional proteins of non-natural composition were prepared through residue-specific incorporation combined with directed evolution.
