Rapid detection of exosomes & exosomal micro RNA biomarkers by electrokinetic concentration for liquid biopsy on chip

Abstract

An electrokinetic concentration device using ion concentration polarization (ICP) is demonstrated for accelerating surface hybridization reaction between exosomal microRNA (miRNA) and morpholinos (MO) as a synthetic oligo capture probe at a nanomolar concentration range within 30 min. as well as to concentrate exosomes from human breast cancer cell lines for rapid detection. The results demonstrate an ICP-based sensing platform that enables significantly shorter detection time at very low concentrations with ease of use and requires no RT-PCR or other conventional enzyme-based amplification methods that could cause artifacts.
